Methylene Blue coloring

Dyes are used in microbiology to help visualize the morphology of bacteria that are practically transparent due to their insignificant measurements.

Simple methylene blue staining makes it easy to see germs, which appear deep blue. Methylene blue is also useful for distinguishing metachromatic granules of Corynebacterium diphtheriae.

Although it is basically used for simple stains, methylene blue is also useful

as a contrast dye in some compound stains (Ziehl-Neelsen).

It is a recommended colorant for young crops (24-36 h).

SIMPLE COLORING WITH METHYLENE BLUE

Method:

  • Use a clean, grease-free slide.
  • Prepare a fine and homogeneous smear. This can be done directly from a culture in liquid medium, from a bacteriological suspension obtained by homogenizing 1-2 colonies in saline solution, or directly from a patient sample.
  • Let dry and fix the smear by placing the slide on top of the flame of a Bunsen burner. Avoid excessive heat.
  • Place the slide in a staining rack.
  • Rinse the slide with methylene blue for 2 minutes.
  • Clean the slide with water using a washing bottle until the water does not turn blue.
  • Allow to drain in a vertical position and dry at room temperature.
  • Visualize with an immersion objective. Put a drop of oil between the smear and the lens.

Storage: Store between 15 and 25ºC
