Reticulocyte staining kit

RETICULOCYTES STAINING AND COUNTING KIT Bright cresil blue and methylene blue stains precipitate with RNA (ribonucleic acid). The latter appearing as blue granules sometimes linked by a reticule.

Since the RNA of the reticulocytes disappears few days after they get into the blood, their counting will serve to know the number of cells released by bone marrow into blood.

The number of reticulocytes is probably the best and easiest indicator of erythropoesis. An increase can be interpreted as a demand for new erythrocytes and proper functioning of the marrow. On the contrary, if the demand decreases or if the marrow does not function

properly, then the number of reticulocytes decreases.

INSTRUCTIONS FOR USE:

Add 3 drops of blood to a tube containing 100 l of stable stain, mix well and incubate for 10- 15 minutes at room temperature. Mix again and make the smear and let dry.

View through an oil immersion objective without posterior stain. In reticulocytes, the precipitated RNA is reticule/reticulated or granular and turns into intense blue. Erythrocytes turn into pale blue or greenish blue. Care must be taken since stain particles that adhere on the surface of the small corpuscles can be misinterpreted.

The percentage of erythrocytes containing reticule or granules remains stable if not exposed to light – a total of at least 1,000 erythrocytes in different parts of the smear must be observed.

Normal results : adults usually have a reticulocyte count ranging between 0,5 and 2 %. Newborns have a count ranging between 2,5 and 6,5 %, decreasing to the adult range after 2 weeks.
